I definitely think at one point they certainly were poised to become something big. That all stopped when their second album (albeit awesome) was a huge departure from what made them big in the first place, and then their third album took forever and was even more of a departure.
The Killers were also part of a movement that was blooming around that time, the sort of post-pop-punk synth/new wave revival. The Killers weren't able to move away from the movement that made them big without losing their notoriety. That's the difference between so many bands and U2, they aren't able to pave their own way and evolve while gaining more and more fans. For most it goes the opposite way.
Almost as ridiculous as when Bono himself said Franz Ferdinand were the next U2.
